The holes that can be seen across the surface of this section are a result of the removal of metal clamps inserted to hold these blocks in place.

Notes

The irregular sizes of the blocks employed for the construction of the Colosseum can be seen here. Even the blocks of the two piers are of different sizes. This is typical of most major monuments in Rome.
